Modulation of MicroRNA Cluster miR-183-96-182 Expression by Epstein-Barr Virus Latent Membrane Protein 1

Abstract: Epstein-Barr virus (EBV)is an oncogenic human herpesvirus involved in the pathogenesis of Burkitt's lymphoma (BL) and various other lymphoproliferative disorders. In BL, EBV protein expression is restricted to EBV nuclear antigen 1 (EBNA1), but small noncoding RNAs such as EBV-encoded small RNAs (EBERs) and microRNAs (miRNAs) can also be detected. miRNAs play major roles in crucial processes such as proliferation, differentiation, and cell death. It has recently become clear that alterations in the expression … Show more

“…We reported a higher serum prevalence of EBNA-1 IgG in MS patients with high levels of miRNA-155. This data was confirmed by other groups that showed the ability of EBV to induce the expression of miR-155 [ 31 ], during latent infection mediated by EBNA1 and LMP, indeed EBV is able to modulate the expression of specific cellular miRNAs, such as miR-155 and miR-146 [ 31 ]. Previous work did not show that natalizumab therapy was able to modify the immune response against EBNA1 in MS patients [ 32 , 33 ].…”

“…During type II and III latency, the γ-herpesvirus EBV selectively downregulates miR-183/96/182 expression [ 114 , 115 , 116 ]. This is mediated by the viral latent membrane protein I (LMP-1), the major oncoprotein of EBV [ 114 ].…”

“…During type II and III latency, the γ-herpesvirus EBV selectively downregulates miR-183/96/182 expression [ 114 , 115 , 116 ]. This is mediated by the viral latent membrane protein I (LMP-1), the major oncoprotein of EBV [ 114 ]. Reduced miR-183/96/182 expression helps push EBV-infected B-lymphocytes and epithelial cells towards a more mesenchymal phenotype, although additional viral and host miRNAs contribute to this phenotypic shift [ 117 , 118 ].…”

“…The use of small RNA cloning and sequencing has been employed in several recent studies to identify up- or down-regulated miRNA upon infection with different viruses ( Motsch et al, 2012 ; Oussaief et al, 2015 ; Chen et al, 2016 ; Lodge et al, 2017 ). However, these studies did not necessarily confirm whether the regulated miRNAs were having pro- or anti-viral roles.…”
