Modulatory Mechanisms of the NLRP3 Inflammasomes in Diabetes

Abstract: The inflammasome is a multiprotein complex that acts to enhance inflammatory responses by promoting the production and secretion of key cytokines. The best-known inflammasome is the NLRP3 (nucleotide-binding oligomerization domain-like receptor [NLR] family pyrin domain-containing 3) inflammasome. The evidence has shown that the NLRP3 inflammasome, IL-1β, thioredoxin-interacting protein (TXNIP), and pyroptosis play vital roles in the development of diabetes. This review summarizes the regulation of type 1 diab… Show more

“…Active inflammasomes can lead to caspase 1-mediated activation of IL-1β [ 38 ]. This effect suggests a potential therapeutic impact of EEO on inflammatory diseases involving the inflammasome, such as type 2 diabetes [ 39 ], inflammatory bowel disease [ 40 ], and atherosclerosis [ 41 ].…”

“…The overexpression of P2X7 receptor exhibits adverse effects on diabetic retinopathy, diabetic neuropathy, and diabetic nephropathy [67][68][69], and the impaired activation of the P2X7 receptor signaling pathway in T1DM may affect the function of osteoblasts and bone health [70,71]. When glucose metabolism is disordered, excessive extracellular ATP level stimulates macrophages in the pancreas and adipose tissues, thus prompting P2X7 receptors to activate NLRP3 inflammasomes [72]. The increased level of NLRP3-dependent IL-1β induces and aggravates T2DM through β cell death and insulin resistance.…”

“…Another way to modulate the innate immune response is to regulate the activation of particular inflammasomes ( Broz and Dixit, 2016 ; Ding et al, 2019 ; Spel and Martinon, 2020 ). Among various types of inflammasomes, nucleotide-binding domain and leucine-rich-repeat-containing family and pyrin domain-containing 3 (NLRP3) is implicated in Alzheimer’s disease and type 2 diabetes upon aberrant activation.…”
