MoErv14 mediates the intracellular transport of cell membrane receptors to govern the appressorial formation and pathogenicity of Magnaporthe oryzae

Abstract: Magnaporthe oryzae causes rice blasts posing serious threats to food security worldwide. During infection, M. oryzae utilizes several transmembrane receptor proteins that sense cell surface cues to induce highly specialized infectious structures called appressoria. However, little is known about the mechanisms of intracellular receptor tracking and their function. Here, we described that disrupting the coat protein complex II (COPII) cargo protein MoErv14 severely affects appressorium formation and pathogenici… Show more

“…Infections occur when fungal spores land on and invade wheat leaves using an appressorium—a specialized infection cell formed during spore germination. The process of the development of appressoria has recently been investigated intensively using genome-wide analysis ( Lv et al., 2022 ; Wang et al., 2022a ; Fan et al., 2023 ; Miguel-Rojas et al., 2023 ; Qian et al., 2023 ; Rogers and Egan, 2023 ). Diverse ecologies have been reported for species of the Magnaporthales, ranging from saprotrophs to cereal pathogens to likely root endophytes ( Luo et al., 2015 ).…”

“…Proteins bound to beads were washed 5 times with GFP washing buffer and eluted with 200 mM glycine (pH 2.5), then neutralized with 1 M Tris (pH 10.4). Input and IP proteins were detected by Western blot using anti-GFP (Engibody Biotechnology, AT0028) and anti-Flag antibodies (Engibody Biotechnology, AT0022), respectively [56].…”
