Molecular Analysis of Anthocyanin Biosynthetic Genes and Control of Flower Coloration by Flavonoid 3′,5′-Hydroxylase (F3′5′H) in Dendrobium moniliforme

Abstract: Dendrobium moniliforme is a native species of Korea. The flower of this species is composed of a reproductive column and white perianths including petals, sepals and lip, but the base of the column bears reddish purple pigment spots. Anthocyanins are major pigments that contribute flower color in Dendrobium. Three key anthocyanin biosynthetic genes encoding dihydroflavonol 4-reductase (DFR), chalcone synthase (CHS), and flavonoid 3',5'-hydroxylase (F3'5'H) were isolated and analyzed for their expression patter… Show more

“…Based on the PGSC database, PG0026477 and PG0000561 were annotated as StAN11, which was homologous to Arabidopsis TTG1. PG0026477b not only interacted with the MYB transcription factor and bHLH1 (PG0012891), but also directly interacted with multiple genes that were involved in the flavonoid biosynthesis pathway, such as ANS (PG0003090, PG0003091, PG0003880, PG0027333, PG0027396, and PG0033567), F3 5 H (PG0000425), DFR (PG0003605), F3H (PG0003563), and FLS (PG1024988) [76][77][78]. PG0026477, annotated as StAN11, plays the same role as the TTG1 gene in anthocyanin synthesis, which was reported to form a MBW complex with the MYB transcription factor and bHLH1.…”

“…Compared with cyanidin-based and pelargonidin-based anthocyanin, the formation of delphinine needed the expression of a special structural gene F3'5'H (Fig 1) [42]. The structural gene F3'5'H had a higher transcript level in blue aleurone than white, which could imply that delphinidin-based anthocyanin was responsible for the aleurone’s blue color in ‘Blue 1’.…”

“…Dendrobium moniliforme "Jindo" (Whang et al 2011) was cultivated under a 16-h light/8-h dark cycle at 24℃ in a greenhouse facility at Chonbuk National University. Leaf and root samples were harvested from young seedlings to isolate total RNA.…”
