A Systematic Meta-Analysis of Global Sarcocystis Infection in Sheep and Goats

Sarcocystosis is an intracellular parasitic disease caused by Sarcocystis spp. that has a worldwide prevalence. Symptoms of the disease include diarrhea and muscle pain. The disease poses a threat to the health of animals. The aim of this review is to investigate the global prevalence of Sarcocystis infection in sheep and goats during 2013–2022. We searched five databases: Web of Science, Science Direct, PubMed, Scopus, and Google Scholar. A total of 36 articles containing 44 datasets met the criteria and were included in the study. The total infection rates of Sarcocystis in sheep and goats were 66.3% (95% CI, 51.79–79.38%) and 52.1% (95% CI, 29.45–74.23%), respectively. It was found that Sarcocystis species tend to have a host species preference. Coinfection of S. tenella and S. arieticanis often occurred in sheep, and goats were frequently infected with S. capracanis. Age and sex were identified as risk factors for Sarcocystis infection in sheep and goats. The infection rates of female and male animals were significantly different, with females having a higher infection rate. Age-adjusted analysis showed that infection rates in animals older than one year were higher than in animals younger than one year. This study unveiled the global distribution of Sarcocystis and sheds light on its transmission in sheep and goats.

IntroductionSarcocystis is a genus of cyst-forming parasites that infest both humans and livestock. Some parasites cause clinical and subclinical diseases in their hosts, resulting in economic losses.MethodsEsophagus, diaphragm, and skeletal muscle from slaughtered sheep and goats were examined macroscopically, microscopically, and ultrastructurally and subjected to DNA analysis.ResultsWe isolated macrocysts of S. gigantea and of S. caprafelis moulei from naturally infected sheep (Ovis aries) and goats (Capra hircus). The macrocyst wall thickness was 18.9 µm in sheep and 15.3 µm in goats, and consisted of an inner Periodic acid Schiff- (PAS) negative primary wall and an outer glycoconjugates containing i.e. PAS-positive secondary wall. The walls inner surface was compartmentalized and filled with bradyzoites. In S. gigantea the bradyzoites were approximently 12.3 x 2.6 µm in size, while in S. caprafelis moulei they were 13.9 x 4.4 µm. Ultrastructurally, both species have nearly identical morphology: cauliflower-like protrusions with numerous microtubules and often dendritic-like filaments, branching from the primary wall. The 18S rRNA gene in S. gigantea was 85.9% identical to that in S. medusiformis and 80.4% to the S. caprafelis moulei gene. The 28S rRNA gene in S. gigantea was 94.6% identical to that in S. medusiformis and 97.3% to the S. caprafelis moulei.ConclusionThis study is the first to (i) detail the ultrastructure of the macrocyst wall of S. caprafelis moulei, (ii) identify S. medusiformis in Iraqi sheep, and (iii) compare the prevalence of macroscopic Sarcocystis at different time periods within the same region. A positive finding was the reduction of macroscopic sarcocystosis occurrences (0.01% in sheep and 0.02% in goats) compared to our previous data from 1992 (4.1%: sheep, 33.6%: goats).

First Molecular Identification of Sarcocystis Buffalonis in Goats Worldwide in Misan Governorate, Iraq

The current study was conducted in Misan Governorate to detect the parasite Sarcocystis spp. by both conventional and molecular methods and to identify the species of this genus in goats as intermediate hosts for multiple species of this parasite. Regarding to the traditional study, the rates of infection with the macroscopic type of Sarcocystis spp. were low and recorded 2% in esophageal samples only in goats. The sizes of macroscopic parasite cysts ranged between 1-10 mm in length and 2-6 mm in width. Infection with the microscopic type of parasite recorded high rates. When comparing the efficiency of the traditional methods used in our study to diagnose the infection of the microscopic type in all of the examined organs isolated from goats, which included the methods of digestion with pepsin, squeezing, trichinoscopy. It was found that the method of digestion with pepsin was the most efficient traditional method for diagnosing the microscopic type of parasite, followed by the trichinoscopy. While the squeezing method recorded the least efficient traditional method for diagnosing Sarcocystis spp. microscopically. The molecular study of Sarcocystis spp. included molecular diagnosis of the parasite's identity at the species level through gene amplification using polymerase chain reaction (PCR) of the target segment of DNA extracted from the parasite samples of goats for 18S rRNA gene by 120 microscopic samples and 4 macroscopic samples. 87.9% of goat samples, showed positive infection with the 18S rRNA gene; In which 100% of the macroscopic and microscopic samples of the esophagus were recorded, and both animals had a positive infection, while the infection rates for the diaphragm, tongue and heart samples were recorded as 90%, 86.7 and 80%, respectively. The results obtained from sequence analysis of 18S rRNA gene for six samples showed that four microscopic samples of goats (OP474017.1, OP474018.1, OP476699.1 and OP474036.1) were closely related to the Malaysian species; S. capracanis with accession number (KR155191.1) with identity score percentages as 99.50%, 99.01%, 99.76% and 99.13%, respectively. Finally, the two macroscopic samples of goats (OP474061.1 and OP474062.1) were identical with the Egyptian species; S. buffalonis, with accession numbers (KU247913.1) and (KU247913.1) respectively, with a matching percentage of 84.39% and 85.64% respectively.
