Molecular and morphological identification of Biomphalaria species from the state of São Paulo, Brazil

Abstract: DNA barcoding and morphological characters were used to identify adult snails belonging to the genus Biomphalaria from 17 municipalities in the state of São Paulo, Brazil. The DNA barcode analysis also included twenty-nine sequences retrieved from GenBank. The final data set of 104 sequences of the mitochondrial cytochrome oxidase I (COI) gene was analyzed for K2P intraspecific and interspecific divergences, through tree-reconstruction methods (Neighbor-Joining, Maximum Likelihood and Bayesian inference), and … Show more

“…Results obtained for species of freshwater and land mollusks which are medically relevant in the neotropics show the importance of combining traditional methods of morphological analysis with molecular methods to identify known species as well as potential cryptic species (Langand et al 1999, Campbell et al 2000, Dejong et al 2003, Wethington et al 2007, Teodoro et al 2010, Tuan et al 2012, Palasio et al 2017, Bezerra et al 2018.…”

“…Identification of Biomphalaria species is challenging because shell features are often highly variable and can even be confused with shells of other genera like Helisoma 44 . Anatomical characteristics, often revealed only by painstaking dissections of specially prepared materials, require experience, patience and practice to discern 13 , 45 , 46 . Nonetheless, reference to both shell and anatomical features in conjunction with provision of an ever-increasing array of molecular data has provided an improved and workable overall systematic framework for Biomphalaria .…”

“…Part of the COI gene (~600 bp) was amplified with the LCO1490/HCO2198 primers (FolMeR et al 1994) (2007). All polymerase chain reactions (PCR) were carried out in a total volume of 50 µl as described in PalaSio et al (2017). The amplified ITS2 fragments were cloned using pGEM-T (Promega).…”
