Molecular character of a phosphatase 2C (PP2C) gene relation to stress tolerance in Arabidopsis thaliana

Abstract: Protein phosphatases type 2C (PP2Cs) from group A, which includes the ABI1/HAB1 and PP2CA branches, are key negative regulators of ABA signaling. HAI-1 gene had been shown to affect both seed and vegetative responses to ABA, which is one of PP2Cs clade A in Arabidopsis thaliana. Transgenic plants containing pHAI-1::GUS (β-glucuronidase) displayed GUS activity existing in the vascular system of leave veins, stems and petioles. Green fluorescent protein fused HAI-1 (HAI-1-GFP) was found in the nucleus through tr… Show more

“…The upregulated genes in all four stress conditions include a NAC TF (AT1G69490 and LOC_Os03G21060), HAI-1 or highly ABA-induced PP2C gene 1 (AT5G59220 and LOC_Os05G38290) and a heavy metal-associated domain containing protein (AT5G52760 and LOC_Os10G38870). Expression of HAI-1 gene was shown to be induced by wound in Arabidopsis [47]. …”

“…S6a), evidence for adaptive evolution is found for a homolog of DEFENSIN J1‐2 , known to inhibit the growth of pathogenic fungi by modifying their morphology or physiology, disrupting or killing exogenous cells (Thomma, Cammue, & Thevissen, 2002). Another candidate gene for adaptive evolution on the D. fuchsii branch is a homolog of HAI1 PHOSPHATASE 2C 78 , which is involved in defence response mechanisms by deposition of callose, a polysaccharide produced as a response to wounding, but also in response to drought and salt stress (Zhang et al., 2013). HAI1 is further involved in the pathway of the endoplasmic reticulum response to an accumulation of unfolded or misfolded proteins on stress (Wahyu Indra Duwi, Lee, & Lee, 2013).…”

“…Under drought stress conditions, ABA accumulates in plants and forms a complex with PYR/PYL. The ABA-PYR/PYL complex inhibits the phosphatase activities of clade A protein phosphatase 2Cs (PP2Cs), leading to the activation of SNF1-related type 2 protein kinases (SnRK2s), which further phosphorylate and activate the downstream substrates such as transcription factors (Schweighofer et al, 2004;Ma et al, 2009;Nakashima et al, 2009;Park et al, 2009;Umezawa et al, 2009;Cutler et al, 2010;Lynch et al, 2012;Rodrigues et al, 2013;Zhang et al, 2013).…”
