Molecular characterisation of Banana mild mosaic virus, a new filamentous virus in Musa spp.

Abstract: Banana mild mosaic virus (BanMMV) is a previously undescribed filamentous virus infecting Musa spp. The complete genome has been sequenced from PCR clones and consists of 7352 nucleotides, encoding five open reading frames (ORFs) of 205, 25.5, 12.4, 8.0 and 26.8 kDa, respectively. BanMMV was most closely related to fovea- and carlaviruses, based on phylogenetic analysis using a portion of the viral replicase. Analysis of other parts of the genome revealed similarities with fovea-, carla- and potexviruses, but … Show more

“…Similar symptoms of chlorotic leaf spotting and fruit deformation are associated with peach infection by Asian prunus viruses (75), while AOPRV is held responsible for a lethal disease of African oil palms (88). None of the putative flexiviruses infecting banana (BanMMV and BanVX) seems to be a serious pathogen (40,113).…”

“…Random RT-PCR was conducted as described by Gambley and Thomas (2001) on purified virus prepared using the modified method of Wakman et al (1995). The heat shock protein 70 homologue (Hsp70h) region of the closterovirid genome was also amplified by RT-PCR using the degenerate primers HSP-P-1 and Tian-P2(M) (Tian et al 1996; water to a final volume of 25 µL.…”

“…AATAAA motif was found completely conserved in two CPMMV isolates from USA (CPMMV-H; AF024628 and CPMMV-M; AF024629; Figure 1) along with 7 HpLV isolates (EF202598, EF202599, EF202600, EF394781, EF394782, EF394783 and EF394784) from China ( Figure 2) and 3 DVS isolates (AJ971469, AJ971470 and AJ971471) from South Korea (Figure 3). This motif is also found in other viruses such as Rice tungro bacilliform virus (RTBV) belonging to caulimoviridae family, where it forms an essential part of the poly (A) signal (Rothnie et al, 2001) and in 3´ untranslated region of ORF5 of Banana mild mosaic virus (genus not assigned) (Gambley and Thomas, 2001). The AATAAA nucleotide motif was absent from multiple sequence alignment of Canada and India.…”
