Molecular characterization, expression analysis, and role of ALDH3B1 in the cellular protection against oxidative stress

Abstract: Aldehyde dehydrogenase (ALDH) enzymes are critical in the detoxification of aldehydes. The human genome contains 19 ALDH genes, mutations in which are the basis of several diseases. The expression, subcellular localization, enzyme kinetics and role of ALDH3B1 against aldehyde- and oxidant-induced cytotoxicity were investigated. ALDH3B1 was purified from Sf9 cells using chromatographic methods and enzyme kinetics were determined spectrophotometrically. ALDH3B1 demonstrated high affinity for hexanal (Km 62 μM), … Show more

“…ALDH3B1 and ALDH3B3 may be responsible for removal of such oxidative stress-induced aldehydes in the plasma membrane. Consistent with this notion, ALDH3B1 protected cells from the cytotoxic, exogenously added aldehydes and oxidants [16].…”

“…ALDH3B1 is also expressed ubiquitously, and its protein levels are high in the kidney, liver, and ovary [15,16]. Similar to ALDH3A2, human ALDH3B1 exhibits activity toward medium-chain to long-chain aldehydes; in contrast to ALDH3A2, its subcellular localization is in the plasma membrane [17].…”

“…ALDH3B1 is expressed in the kidney, liver, and brain and has been shown active in vitro toward C6 to C9 aldehydes [12]; however, its activity toward ≥C10 aldehydes is currently unknown. In addition, although ALDH3B1 is suggested to be localized in the plasma membrane and cytosol [13,14], the precise intracellular localization still remains to be elucidated. Unlike other ALDH3 family members, ALDH3B2 has so far not been well characterized.…”
