Molecular Characterization, Expression Patterns, and Association Analysis with Carcass Traits of Porcine USF1 Gene

Abstract: The upstream stimulatory factor 1 (USF1) is a transcription factor controlling expression of several genes involved in lipid and glucose homeostasis. In this study, two isoforms of the porcine USF1 gene were detected by reverse transcription polymerase chain reaction (RT-PCR), termed USF1 wild-type (wt) and USF1/CD, both of them contain a helix-loop-helix leucine zipper (HLH-LZ) conserved domain. Tissue distribution analysis showed that the two transcripts of porcine USF1 gene were ubiquitously expressed in al… Show more

“…As an important regulatory factor, USF has highly conserved bHLH-LZ domain and binds to consensus sequence (CANNTG) of E-box to further regulate the transcription process of different proteins (Wu et al, 2013; Lupp et al, 2014). USF1 was reported to regulate the expression of IL-10 in glioma related microglia: inhibition of USF1 expression resulted in the up-regulation of IL-10 expression (Zhang et al, 2007).…”

“…The human upstream transcription factor 1 (USF1) gene is located in the q22.3 region of chromosome 1. USF1 contains an HLH-LZ structure (helix-loop-helix leucine zipper) that can bind to the E-box of various gene promoter regions 12 and is involved in regulating genes related to lipid and carbohydrate homeostasis in the human body 13 . During DNA damage, USF1 can stabilize the p53 protein and briefly halt the cell cycle 14 .…”

“…Upstream stimulating factor 1 (USF1) is a critical component of the basic helix-loop-helix leucine zipper (bHLH-LZ) family of transcription factors. 35 The HLH-LZ structure contained in USF1 can combine with the E-box of many gene promoter regions. 36 Therefore, USF1 involves in the transcription process of numerous proteins and works as an important regulatory factor in plenty of diseases, including cancer.…”
