Molecular Characterization of a Strain ofSquash Leaf Curl China Virusfrom the Philippines

Abstract: The complete nucleotide sequence of infectious cloned DNA components (A and B) of the causal agent of squash leaf curl disease in the Philippines was determined. DNA‐A and DNA‐B comprise 2739 and 2705 nucleotides, respectively; the common region is 174 bases in length. Five ORFs were found in DNA‐A and two in DNA‐B. Partial dimeric clones containing DNA‐A and DNA‐B, constructed in a binary vector and transformed into Agrobacterium tumefaciens, induced systemic infection in agro‐inoculated pumpkin plants (Cucur… Show more

“…Since then, the diseases caused by begomoviruses have been reported in bitter gourd, chayote, cucumber, muskmelon, pumpkin and sponge gourd [17,20,22,30,34]. Many different begomoviruses infecting cucurbits have been reported from other countries such as Squash leaf curl Yunnan virus from China [39], Melon chlorotic leaf curl virus from Guatemala [3] Chayote yellow mosaic virus from Nigeria [33], ToL-CNDV from Thailand [26,27], Watermelon chlorotic curly stunt virus from Sudan and Iran [13], Squash leaf curl Philippines virus from the Philippines [14], Cucurbit leaf curl virus [2], Cucurbit leaf crumple virus [9], Squash leaf curl virus [8,16] and Squash mild leaf curl virus [4] from USA, and Luffa yellow mosaic virus from Vietnam [24].…”

“…In the Eastern Hemisphere, Watermelon chlorotic stunt virus has a native range spanning Sudan, southern Arabia, and Iran (23,33). (25) occurs in China, the Philippines (34), and Vietnam (34), whereas Squash leaf curl Yunnan virus has been reported only in southern China (54). Loofa yellow mosaic virus is host and geographically restricted and is endemic in southern Vietnam (43).…”

“…For further confirmation of begomovirus infectivity, Southern hybridization was performed according to the method of Kon et al [9]. Cloned DNA-A of Papaya leaf curl virus was used to develop the radiolabeled probes as a general probe for begomoviruses.…”
