Molecular characterization of a Xylanase-producing fungus isolated from fouled soil

Abstract: Xylanase (EC 3. 2. 1. 8), hydrolyzes xylo-oligosaccharides into D-xylose and required for complete hydrolysis of native cellulose and biomass conversion. It has broad range of applications in the pulp and paper, pharmaceutical and Agri-food industries. Fifty fungal species were isolated from the fouled soil around an oil refinery and screened for the production of xylanase enzyme by enrichment culture techniques. The isolated fungal strain was identified as Hypocrea lixii SS1 based on the results of biochemica… Show more

“…For cellulases, molecular weight sizes of 25-50 kDa were reported in Trichoderma reesei and Phanerochaete (Nayebyazdi et al, 2012), 20-45 kDa in Aspergillus niger (Ncube et al, 2012), 25-100 kDa in endophytes (Elisashvili et al, 2015) and 30-250 kDa in most fungi (Kuhad et al, 2011;Li et al, 2011;Liming and Xueliang, 2004;Santa-Rosa et al, 2018;Yasmin et al, 2013;Ritter et al, 2013;Zhang and Zhang, 2013). For xylanases, molecular weight sizes of 20-50 kDa were reported in endophytes (Polizeli et al, 2005), 18-52 in Aspergillus aculeatus (Fujimoto et al, 1995), 29 kDa in Hypocrea lixii (Sakthiselvan et al, 2014), 19 kDa in Aspergillus fumigatus (Silva et al, 1999) and 45-70 kDa in Neocallimastix frontalis (de Segura and Fevre, 1993).…”

“…The extent of germination of most spores is influenced by media components and environmental factors such as temperature, pH, relative humidity, etc. (Sakthiselvan et al 2014;Colin et al 2013). In the present work, the experiment was carried out with optimized seed media which was developed in the laboratory and the effect of pH and temperature on germination of spore with respect to time was critically evaluated.…”

“…Por otra parte, los tratamientos de T. harzianum (N) y Azotobacter sp. (B) inhibieron el crecimiento de las mismas, sin embargo, Sakthiselvan et al (2014) han sugerido que T. harzianum (N) puede favorecer el crecimiento vegetal, y cuando se mezcla con carbón de bajo rango (CBR) generan un efecto positivo sobre algunas propiedades químicas del suelo aumentando la solubilización de contaminantes y la capacidad de absorción de los mismos.…”
