Molecular characterization of cathepsin L cDNA and its expression during oogenesis and embryogenesis in the oriental river prawn Macrobrachium nipponense (Palaemonidae)

Abstract: ABSTRACT. We identified the cDNA sequence of cathepsin L (CatL) in Macrobrachium nipponense, designated as MnCatL, for the first time. The MnCatL cDNA, isolated from the ovary, was 1710 bp in length, containing a 31-bp 5'-untranslated region, a 650-bp 3'-untranslated region, and an open reading frame of 1029 bp, encoding a 342-amino acid polypeptide with a predicted molecular mass of 37.7 kDa. The polypeptide is composed of an 18-amino acid signal peptide, a 106-amino acid propeptide and a 218-amino acid matur… Show more

“…In the current study, we found high occurrence of Vg and VgR in ovary of P. clarkii , and RT-PCR demonstrated that the expression level in ovary is significantly higher than in testis which provided some clues to further elucidate the function to ovary development of P. clarkii ( Table S1 ). Cathepsins belong to a family of proteases that cleave other proteins and are ubiquitously present in almost all organisms [41] , and involved in ovary maturation and embryo development [14] , [42] . There are two kind of cathepsins: cathepsin C and L in our study, and were largely expressed in ovary libraries.…”

“…Although PTGR1 is involved in the ovary development in the shrimp Penaeus monodon [80], it was not preferentially expressed in the ovaries of Palaemon serratus . Cathepsins have been also related with ovarian development in some crustaceans [22, 81, 82]. Cathepsin D is a needed protein for the formation of the yolk in vertebrates [83] and its gene was the only cathepsin gene up-regulated in the ovary of P. serratus .…”

“…The activity of carboxypeptidases А and В, as well as that of leucine aminopeptidase (Dendinger, 1987;Glass & Stark, 1995) was detected also. In various shrimp tissues cDNA coding cathepsin L (Duan et al, 2013;Zhao et al, 2013), cathepsin B (Stephens et al, 2012;Li et al, 2013) and cathepsin С (Wang et al, 2012) were revealed. These data suggest that chironomid larvae may possess a wide range of peptidases.…”
