Molecular characterization of Gastrodiscoides hominis (Platyhelminthes: Trematoda: Digenea) inferred from ITS rDNA sequence analysis

Abstract: Gastrodiscoides hominis (Digenea: Paramphistomata: Gastrodiscidae) is an amphistomid intestinal fluke of pigs causing gastrodiscoidiosis. With the use of molecular tools assisting the conventional diagnostic procedures, we aimed at finding out molecular characterization of G. hominis using PCR amplifications of rDNA ITS (1, 2) sequences. The sequences obtained (GenBank accession numbers EF027096, EF027097, EF027098, EU887294, and EU887295) were compared with available sequences of other digenean parasites, par… Show more

“…Previous studies have used the easily obtained ITS2 sequence as a molecular marker to distinguish among paramphistomoid species (Itagaki et al 2003 ; Rinaldi et al 2005 ; Goswami et al 2009 ; Lotfy et al 2010 ; Sanabria et al 2011 ; Ichikawa et al 2013 ; Shylla et al 2013 ; Ghatani et al 2014 ; Dube et al 2015 ). ITS2 is helpful for distinguishing paramphistomoid genera and differentiating more divergent species within a genus (Rinaldi et al 2005 ; Ghatani et al 2012 ).…”

“…Because of such problems, the development of alternative approaches to better delineate species in this group is needed. PCR-based techniques providing rDNA ITS2 sequences have proven to be a reliable tool to identify digenean species and to recover their phylogenetic relationships (Blair et al, 1999;Itagaki et al, 2003;Reinitz et al, 2007;Lotfy et al, 2008;Goswami et al, 2009;Van Van et al, 2009). Moreover, ITS2 has been found to be a useful marker for species identification of paramphistomes (Itagaki et al, 2003;Rinaldi et al, 2005;Goswami et al, 2009).…”

“…In recent years, several molecular tools including sequence data related to conserved domains have unveiled their utility in molecular identification, taxonomy, systematics and phylogeny of floral, faunal and microbial species, too (Huang et al 2006;Prasad et al 2009a, b;Wiemers et al 2009;Yao et al 2010;Sharma et al 2012). Since Woese and Fox (1977), the nuclear ribosomal repeat cistrons have also been widely implied for parasites (Littlewood and Olson 2001;Waeschenbach et al 2007;Jyrwa et al 2009;Goswami et al 2009;Tandon et al 2010;Prasad et al 2011;Ghatani et al 2012). Molecular phylogeny has advanced our understanding of comparative studies in basic and applied biology and more generally of their position within the phylum Platyhelminthes (Brooks and Hoberg 2001;Tandon et al 2010).…”
