Molecular Characterization of Human Lymph Node Stromal Cells During the Earliest Phases of Rheumatoid Arthritis

Abstract: Rheumatoid arthritis (RA) is a progressive, destructive autoimmune arthritis. Break of tolerance and formation of autoantibodies occur years before arthritis. Adaptive immunity is initiated in lymphoid tissue where lymph node stromal cells (LNSCs) play a crucial role in shaping the immune response and maintaining peripheral tolerance. Here we performed the first epigenomic characterization of LNSCs during health and early RA, by analyzing their transcriptome and DNA methylome in LNSCs isolated from lymph node … Show more

“…As LNs are the epicenter for Tcell activation and differentiated and activated T cells play a key role in PsA, it will be of great relevance to study those cellular responses in LNs, especially during the onset of PsA. Our research group has recently successfully initiated LN tissue sampling (116)(117)(118)(119) of patients with inflammatory arthritis, RArisk individuals and healthy controls (116)(117)(118)(119). We have found altered frequencies of immune cells in LN biopsies of patients with RA and RA-risk individuals (117,119).…”

“…DNA methylation changes in these pathway showed similar pattern with human cancer implicates synovial hyperplasia and invasion maybe shared common underlying mechanism with human cancer metastasis . What's more, extracellular matrix (ECM), cholesterol biosynthesis and immune system pathways also significantly enriched in RA high-risk individuals and RA patients (Karouzakis et al, 2019). Genome-wide methylation levels change was also found in both T and B lymphocytes (Glossop et al, 2013;Rhead et al, 2017;Guderud et al, 2020).…”

“…DNA methylation changes in these pathways show a similar pattern with human cancers, implicating that synovial hyperplasia and invasion may be a shared underlying mechanism with human cancer metastasis (Wang et al, 2015). Moreover, extracellular matrix (ECM), cholesterol biosynthesis, and immune system pathways are also significantly enriched in RA high-risk individuals, indicating that DNA methylation signals may be useful in early diagnosis or risk evaluation of RA (Karouzakis et al, 2019).…”
