Molecular Characterization of Parthenogenic Fasciola sp. in Korea on the Basis of DNA Sequences of Ribosomal ITS1 and Mitochondrial NDI Gene

Abstract: ABSTRACT. Nucleotide sequences of ribosomal internal transcribed spacer (ITS1) and mitochondrial NADH dehydrogenase I (NDI) gene were analyzed to genetically characterize aspermic Fasciola forms in Korea. From the difference in ITS1 sequences, Korean flukes were divided into 3 haplotypes represented by Kor1, Kor2 and Kor1/2, which had nucleotides identical to F. hepatica, F. gigantica and those overlapped between the two species, respectively. NDI sequences also showed that Korean flukes could be classified in… Show more

“…using high pure PCR template preparation kit (DynabioÂź, Takapouzist, Iran), according to the manufacturer's instructions, and stored at -20°C until use. The ITS1 region, as a nuclear marker, was amplified with primers named ITS1-F and ITS1-R and fragments of mitochondrial target (ND1) by polymerase chain reaction (PCR) (18,19). Total volume of the reaction was 40 ”L containing 4 ”L DNA template, 14 ”L distilled water, 10 pmol of each primer, and 20 ”L master mix (ampliconÂź).…”

“…Aspermic Fasciola species are distributed throughout Asian countries, including Japan [7,12], Korea [10], China [15], Vietnam [11], and Myanmar [8]. Particularly in the former 2 countries, all of the Fasciola flukes analyzed were aspermic [7,10,12,18].…”

“…Although Fasciola specimens in Japan have been considered to be of the same origin as that of aspermic Fasciola specimens in Korea, China and Vietnam [7][8][9]16], the frequency of DNA type combinations in Japanese specimens apparently differs from that in those three countries (e.g., the combination of Fsp1/2 in ITS1 and Fsp2 in ND1 is conspic- Table 1. ), M: 100 bp DNA ladder.…”
