Panton–Valentine leukocidin gene is produced by Staphylococcus aureus, and methicillin-resistant Staphylococcus aureus isolates as a pore-forming toxin is largely responsible for skin and soft tissue illnesses. MRSA produces PVL toxins through lukS and lukF proteins causing tissue necrosis by damaging membrane of the defense cells. Presence of PVL toxin was tested from the 54 S. aureus clinical isolates obtained from Thika and Kiambu Level 5 Hospitals, in Kiambu County, Kenya, by Geno Type® MRSA assay (Hain Life Science, Nehren, Germany). DNA was isolated from freshly harvested bacterial cultures by spin column using Geno Type DNA isolation kit. The detection of PVL toxins was performed by amplification of genomic DNA and by reverse hybridization that identifies PVL genes using Geno Type MRSA kit. Out of 138 samples that were collected from patients in Kiambu County, 54 S. aureus isolates were obtained, of which 14 (25.9%; 95% CI = 11.9–38.9) samples had PVL toxins. The isolates that were obtained from the female patients had a higher PVL toxin prevalence of 35.7%, while the isolates collected from the male patients had a lower prevalence of 15.4% (P=0.09). The pediatrics department had the highest PVL gene prevalence compared to outpatient department and surgical units (P=0.08). However, the age groups of patients and the hospital attended by patients showed no significant difference in terms of PVL gene prevalence (P=0.26). Therefore, the patients' gender and hospital units were not significantly associated with PVL gene prevalence (P=0.08). This study shows that PVL positive isolates occur in the sampled hospitals in the county and female as well as children must be taken into consideration among patients with wound infections when isolating S. aureus.

Background: Acinetobacter baumannii is capable of forming biofilms that may be responsible for the survival of this pathogen in the hospital environment as well as antibiotic resistance. Objectives: In this study, considering the importance of genes bap, blaPER-1, and csuE in the formation of biofilms and resistance to antimicrobial drugs, we aimed to investigate the frequency of these genes and also the relationship between these genes and the biofilm formation. Methods: One hundred and eighteen clinical strains of the A. baumannii were collected and identified using standard microbiological methods. Antibiotic susceptibility was evaluated by microdilution broth and disk diffusion methods according to the Clinical and Laboratory Standards Institute (CLSI). Biofilm formation assay was performed by microtiter plate method. Then the bap, blaPER-1, and csuE genes were detected by PCR. Results: The rate of XDR and MDR were 16.1% and 83.9%, respectively. Moreover, 9 (7.6%) isolates were resistant to colistin. The results of biofilm formation revealed that 32 (27.1%), 33 (28.0%), 37 (31.4%), and 16 (13.6%) of the isolates had non-biofilm, weak, moderate, and strong activities, respectively. The association between the formation of biofilm and amikacin resistance was found (P < 0.05). In the isolates, the frequencies of bap, blaPER-1, and csuE genes were 70.3%, 54.2%, and 93.2%, respectively. Statistical analysis showed a significant correlation between the frequency of blaPER-1 and bap genes and the ability to form biofilms (P < 0.05). Conclusions:This study shows the high tendency among the clinical isolates of A. baumannii to form a biofilm. It also shows the correlation between the presence of blaPER-1 and bap genes with the capacity of biofilm formation. Moreover, the majority (92.4%) of the A. baumannii isolates from Isfahan were susceptible to colistin. Therefore, providing new and effective strategies is essential for the prevention and treatment of infections caused by biofilm-forming A. baumannii strains.

Acinetobacter baumannii has gained attention for years as a significant clinical problem due to the increase of antibiotic-resistant strain. Indeed, A. baumannii OmpA is one of the highly conserved membrane proteins among Gram-negative bacteria that has multiple roles in interacting with the host during infection, thereby representing an effective target for the development of novel antibacterial or vaccination therapies. Nowadays, finding suitable epitope-specific antigens inside the conserved proteins such as OmpA is a promising method for successful vaccination programs. Therefore, in the present study, the coding sequence of the 240 to 356 amino acid residues of A. baumannii OmpA (AbOmpA240-356) was cloned into the vector pET-28a and purified using nickel affinity chromatography. In addition, the anti-His tag antibody is used to validate its production. This system of protein expression and purification may be useful for further characterization of AbOmpA240-356 protein fraction. Therefore, this study can lead to the introduction of suitable candidates for the development of an effective vaccine based on OmpA against this bacterium for further analysis.
