“…The procedures used to engineer and express Ig-OVA are similar to those described for Ig-PLP1 (20). In brief, the 91A 3 V H gene (24) was subcloned into the EcoRI site of the pUC19 plasmid and used as template DNA in PCR mutagenesis reactions to generate 91A 3 V H fragments carrying OVA 323-339 nucleotide sequence in place of the D segment within complementarity determining region 3 region. The resulting 91A 3 V H -OVA fragment was then analyzed by nucleotide sequencing to ensure that the D segment was deleted and a nucleotide sequence that will encode full OVA peptide was inserted instead.…”
