Molecular Cloning and Expression of Ferritin mRNA in Heavy Metal-PoisonedXenopus laevisCells

Abstract: In a search for genes transcriptionally regulated by metal ions, we have isolated a Xenopus laevis ferritin cDNA clone, XL2-17, from cadmium-poisoned XL2 cells. The large size of the corresponding ferritin mRNA (1.4 kb) is due to the presence of a 629-nucleotide 5'-untranslated region. The Xenopus ferritin sequence is highly isologous with other vertebrate ferritins. In particular, there is a complete sequence identity for the iron-responsive element (IRE) located in the 5'-untranslated region in both XL2-17 a… Show more

“…Semiquantitative RT-PCR revealed that the transcriptional level of AlFER was the highest in the intestine, followed by the respiratory tree, perisoma and muscle strips, with no mRNA detected in the gonad. It is noteworthy that the intestine of A. leucoprocta is always fi lled with mud and therefore is potentially challenged by bacteria and metal ions that may cause up-regulation of ferritin expression (Muller et al, 1991;Zhang et al, 2006). The absence of ferritin in gonad coincides with reports on Haliotis rufescens and H .…”

“…In contrast, the heme‐free ferritin encoded by cj0612c involved in intracellular iron storage was upregulated. Ferritin is involved in the primary detoxification response to heavy metals including Cd 2+ in Xenopus laevis cells [53]. The principal function of ferritins is to store iron inside cells in the ferric form; a secondary function could be detoxification of iron or protection against O 2 and its reactive products.…”

“…1983, Goudard et al 1991, Milcent et al 1996. Thus, some authors have suggested that ferritin may be a general metal detoxicant protein carrier (Joshi & Zimmerman 1988, Joshi et al 1989, Muller et al 1991. Taking into consideration these observations, the binding of 2'0Po by ferritin in fish liver is, therefore, not surprising.…”
