Molecular cloning and sequence analysis of putative chicken prolactin cDNA

Abstract: A cDNA library was prepared from mRNA isolated from anterior pituitary glands of incubating bantam hens, in which prolactin mRNA levels were predicted to be very high. Nine clones, representing abundant mRNA species, were identified and shown to contain homologous sequences. Two clones, of 871 bp and 580 bp, were analysed by DNA sequencing. The shorter clone was found to be a truncated cDNA product but otherwise identical to the longer clone. The 871 bp cDNA, PRL101, contains an open reading frame capable of e… Show more

“…Gaps have been introduced to obtain maximal homology. Sequences for man (Cooke, Coit, Shine et al 1981), pig (Kato et al 1990), ox (Sasavage, Nilson, Holowitz & Rottman, 1982), rat (Cooke, Coit, Weiner et al 1980), mouse (Linzer & Talamantes, 1985), chicken (Hanks, Alonzi, Sharp & Sang, 1989), salmon (Song, Trinh, Hew et al 1988) and bullfrog (present study) are shown.…”

“…Because nucleotide sequences are higly conserved between chicken and quail, chicken and quail probes are widely used for both species in methods such as Northern hybridization, Southern hybridization, in situ hybridization, chromosomal mapping using FISH, and microarrays (17,(35)(36)(37)(38)(39)(40). Therefore, we used chicken DNA sequences for in situ hybridization with 33 P-labeled (17).…”
