Molecular demultiplexer as a terminator automaton

Abstract: Molecular logic gates are expected to play an important role on the way to information processing therapeutic agents, especially considering the wide variety of physical and chemical responses that they can elicit in response to the inputs applied. Here, we show that a 1:2 demultiplexer based on a Zn2+-terpyridine-Bodipy conjugate with a quenched fluorescent emission, is efficient in photosensitized singlet oxygen generation as inferred from trap compound experiments and cell culture data. However, once the si… Show more

“…An example of such an advanced photodynamic treatment of cancer cells was recently demonstrated by Akkaya and co-workers using dyad 80. 98 The Zn(II)-terpyridyl fragment in this system plays the role of electron acceptor, promoting PET from the BODIPY subunit ( Figure 19a). The dyad generates singlet oxygen ( = 0.1) and triggers apoptosis in cancer cells under light irradiation.…”

“…The always‐on property of PSs results in high background fluorescence and may reduce the treatment specificity . This problem can be efficiently overcome by designing a PS that is activated in the tumor microenvironment, which can be achieved using the concept of molecular logic gates . In this case, only inputs to the tumor microenvironment such as low pH, thiols, or cancer‐related enzymes together with light irradiation can lead to the output of toxic ROS.…”

“…Once a cell starts to become apoptotic, many distinct biochemical changes occur. A particularly noteworthy theranostic construct was recently reported by Turan et al . Using molecular logic gates, they exploited their deep understanding of PeT quenching to develop a Zn 2+ ‐terpyridine‐BODIPY conjugate that demonstrates self‐driven photosensitization turn‐off with simultaneous fluorescence turn‐on following PDT induction of apoptosis.…”
