Molecular Design of Photosynthesis-Elevated Chloroplasts for Mass Accumulation of a Foreign Protein

Abstract: In order to increase production of a useful protein by the chloroplast transformation technique, it seems to be necessary to determine the upper limit for the accumulation of a biologically active foreign protein in chloroplasts and then improve photosynthetic capacity and plant productivity. Here we show that the stromal fractions of tobacco chloroplasts could accommodate an additional 200-260 mg ml(-1) of green fluorescent protein in the stroma without any inhibition of gas exchange under various light inten… Show more

“…Moreover, using of a promoter driven by a naturally available stimulus is eco-friendly and does not change the circadian expression of genes of microalgae [15]. The P psbA1 promoter from other sources has earlier been used for expression of desired genes in Anabaena [16] and in other cyanobacteria [17] and plants [18]. Our findings indicate that the pAnFP vector bearing heterologous genes can be successfully expressed in E. coli and be used for transformation of cyanobacteria such as Anabaena or Nostoc strains in the future.…”

“…On the basis of these results, transplastomic tobacco (Yabuta et al 2008) and lettuce plants (Ichikawa et al 2010) expressing cyanobacterial fructose-1,6/ sedoheptulose-1,7-bisphosphatase (FBP/SBPase) in the chloroplast have recently been generated, and both exhibited increased productivity. In tobacco plants, expression by three different plastid promoters caused a minimum of 2-or 3-fold increase in the enzyme activity, which resulted in up to 1.8-fold increase of the dry matter when compared to the nontransformed plants.…”

“…In tobacco plants, expression by three different plastid promoters caused a minimum of 2-or 3-fold increase in the enzyme activity, which resulted in up to 1.8-fold increase of the dry matter when compared to the nontransformed plants. This increase was sufficient to improve productivity and photosynthesis without representing a strong metabolic burden to the plastid and potentially enabled the simultaneous high-level expression of other foreign genes in the plastids (Yabuta et al 2008). In addition, the hexose, sucrose, and starch contents of the stem, root, and different leaves were in general higher in transplastomic plants than in non-transformed plants (at least during distinct stages of the diurnal cycle) (Yabuta et al 2008).…”

“…Tobacco leaf accumulated green fluorescent protein of 30% of total soluble proteins when its gene was introduced into the chloroplast genome. 8 Chloroplast transformation takes place by particle bombardment of gold particle coated by vectors against green organelle, and regeneration of transformed plants successfully takes place in several plants, such as tobacco, potato and lettuce.…”
