Molecular Detection of Theileria Species in Sheep from Northern China

Abstract: ABSTRACT. Ovine theileriosis is a tick-borne disease that restricts the development of small ruminant husbandry in northern China. In this study, we report on a molecular epidemiological survey of ovine Theileria spp. in 198 blood samples taken from sheep in northern China. The DNA samples were screened by a nested polymerase chain reaction (PCR) targeting the 18S rRNA gene of ovine Theileria spp. The prevalence of ovine Theileria spp. in Yanji, Nongan, Longjing, Toudao and Jinchang was 80%, 40%, 37%, 24% and … Show more

“…Specific primers targeting B. bovis spherical body protein-4 (SBP-4) , B. bigemina rhoptry-associated protein-1a (RAP-1a), Theileria spp. 18S rRNA and A. marginale major surface protein 5 (Msp5) genes were used to amplify the respective genes by using previously described nPCRs [ 20 22 ]. Initial PCR amplifications were done in a 10 μl-reaction mixture having 1 μl of DNA template, 1 μl (10 μM) of each primers, 1μlof 10x Ex buffer, 1 μl of dNTP (200 μM each), 0.1 μl of Ex Taq polymerase (Takara, Japan) and 4.9 μl of double distilled water.…”

“…Theileria species that infect bovines include T. annulata , T. parva , T. mutans , T. orientalis complex ( orientalis/sergenti/buffeli ), T. tarurotragi , T. velifera , T. sinensis and Theileria sp. Yokoyama, a newly discovered Theileria species closely related to T. annulata [ 8 , 9 , 10 , 11 , 12 , 13 ]. The geographic distribution of Theileria species is reported in Table 1 , along with the disease names they cause and the vector tick species.…”

“…These asymptomatic carriers show low levels of parasitaemia which is usually difficult to detect by microscopy [10]. Therefore, molecular techniques are usually more accurate in diagnosing subclinical infections [11,12]. Trypanosomosis (Surra) is a mechanically transmitted disease of cattle and horses.…”
