Molecular ecology of Triatoma dimidiata in southern Belize reveals risk for human infection and the local differentiation of Trypanosoma cruzi parasites

Abstract: In Belize, the main vector for Trypanosoma cruzi, the agent of Chagas disease, is Triatoma dimidiata, but transmission cycles and the risk for human infection are unclear. Therefore, the aim of this study was to identify T. dimidiata blood feeding sources and its parasite and microbial diversity, in order to reconstruct T. cruzi parasite transmission ecology in southern Belize. Methods: A metabarcoding approach based on deep sequencing of markers was used for bug taxonomy, blood meal sources, T. cruzi genotype… Show more

“…Amplicon deep sequencing fills the need for a powerful method that detects not only the most recent/abundant host DNA, but also older and partially degraded bloodmeals as well. This method is becoming increasingly used in the detection of bloodmeals from triatomines yielding multiple host information from individual bugs allowing comparison of triatomines from different habitats, understanding their behaviors and controlling the spread of Chagas disease 13 15 , 52 .…”

“…After mapping using all the DTUs as reference, a total of 89,746 sequences of good quality mapped only to the TcIV DTU. The phylogenetic analysis indicated four unique TcIV min-exon sequence haplotypes, which were most closely related to the TcIV T. cruzi sequences identified from T. dimidiata vectors previously sequenced from southern Belize 13 as well as the acute Chagas disease pediatric case-patient 18 represented in this study. Yet they were distinct from the T. cruzi TcIV reference, CanIII, from Brazil (Fig.…”

“…and T. huehuetenanguensis (formerly published as clade III T. dimidiata s.l.) were thought to be predominately sylvatic in nature, but able to seasonally infest houses, and hybrids have also been identified 12 , 13 , 15 . While our findings from this small sampling are not robust enough to draw significant conclusions about vector ecology of this potential new species, we made some observations of note.…”

“…A general trend in field-caught bugs is that nymphs have a highly diverse microbiota, which often differs from that of adult bugs, suggesting different metabolic functions and interactions. This is the case for Triatoma protracta [17], Triatoma reticularia [17], Triatoma rubida [17], Triatoma dimidiata [18,19], and Rhodnius prolixus [20]. On the other hand, in Triatoma sordida the adult microbiota is more diverse compared to nymphs, with unique bacteria being incorporated from the 4 th instar stage onwards [21].…”
