Molecular function of the dual‐start motif in the λ S holin

Abstract: SummaryThe l S gene represents the prototype of holin genes with a dual-start motif, which leads to the synthesis of two polypeptides, S105 and S107. They differ at their N-terminus by only two amino acids, Met-1 and Lys-2, at the beginning of the longer product. Despite the minor difference, the two proteins have opposing functions in lysis, with protein S107 being an inhibitor and protein S105 being an effector of`hole formation' in the inner membrane. Here, we have studied the molecular mechanism underlying… Show more

“…Physiological and biochemical studies indicated that this charge prevents movement of the N terminus, including TMD1, of nascent S107 through the membrane, so that the antiholin has only two TMDs (Fig. 1B) (13). This form of S107 dimerizes with and inhibits S105 (14).…”

“…Cooperative action of the inhibitor and effector would result in proper scheduling of cell lysis ("lysis clock"). Major progress in understanding the molecular mechanism underlying regulation of lysis by the dual start motif has been reported recently for the lambda encoded holin (88). Gene 14 of B103 has similar features to that of 29, even though the second amino acid residue of the predicted inhibitor species is an Asp instead of a Lys.…”

“…Genes containing dual translational initiation signals and thus generating signal sequences with various lengths have been described previously. Taguchi et al (1991) suggested that dual starts could affect gene expression and Graschopf & Blasi (1999) reported that the different starts result in mature proteins of different functions. In L. reuteri, the gene encoding the mucus-binding protein Mub has two possible start motifs generating signal peptides of different lengths (Roos & Jonsson, 2002).…”
