Molecular Identification of Several Orchid Species Based on OPA10 and OPA18 RAPD Marker

Abstract: The purpose of this study was to determine the genetic characteristics of nine orchids plants collected by Biology Gardens of Mathematics and Science Faculty of UNY based on OPA10 and OPA18 markers, to detect their genetic relationship, and determining the exact RAPD markers for identification of intraspecies and interspecies orchids. The analysis was carried out by isolating DNA from the leaves of 9 species of orchids (Rhynchostylis retusa, Vanda tricolor, Dendrobium aphyllum, Dendrobium fimbriatum, Dendrobiu… Show more

“…Sebanyak sekitar 5.000 spesies anggrek dapat ditemukan di Indonesia. Anggrek menjadi populer karena memiliki bunga yang indah, harum, serta bentuk dan coraknya yang menarik (Kurniawati et al 2019). Secara holistik, tanaman ini berperan penting dalam berbagai aspek kehidupan mulai dari industri tanaman ornamental hingga studi evolusi.…”

“…Furthermore, DNA bands measuring 450 bp in D. secundum, and D. bracteosum had the same band size. Kurniawati et al (2019) used RAPD as a genetic marker to characterize several Dendrobium orchids. Based on RAPD results using RAPD OPA10 and OPA18, D. anosmum gigantea alba and D. anosmum gigantea showed different genetic characteristics but some DNA fragments had the same size (918 bp and 492 bp).…”

“…Hence, those constraints affect the position of species in the phylogenetic tree (Minasiewicz et al 2018;Yun et al 2020). The discovery of new species from crosses also regularly changes the grouping and name of an orchid species (Kurniawati et al 2019). The genetic distance calculated using molecular marker data may be used to predict the existence of sexual incompatibility among members of the Aeridinae subtribe.…”

“…The presence of intergeneric hybrids within the Aeridinae subtribe has contributed to the complexity of the biological concept of species in orchids (Surveswaran et al 2018) and eventually revised the modern species concept (Aung and Jin 2018;Zhou et al 2021). The new species discovery from intergeneric and interspecific hybrids results in regular changes in the grouping and naming of orchid species (Kurniawati et al 2019).…”
