Molecular imaging of ectopic metabotropic glutamate 1 receptor in melanoma with a positron emission tomography radioprobe18F-FITM

Abstract: Oncoimaging using positron emission tomography (PET) with a specific radioprobe would facilitate individualized cancer management. Evidence indicates that ectopically expressed metabotropic glutamate 1 (mGlu1) receptor independently induces melanocyte carcinogenesis, and it is therefore becoming an important target for personalized diagnosis and treatment strategies for melanomas. Here, we report the development of an oncoprotein-based PET imaging platform in melanomas for noninvasive visualization and quantif… Show more

“…Accumulation of 4-[ 18 F]fluoro-N- [4-[6-(isopropylamino)pyrimidin-4-yl]-1,3-thiazol-2-yl]-N-methylbenzamide ([ 18 F] FITM) in metabotropic glutamate 1-positive melanoma lesions was shown in tumor-bearing mice with favorable signal-to-noise ratios. 138 Other groups pursued the concept of theranostics. Qin et al 139 proposed a tracer binding at the human copper transporter 1 (CTR1), which is overexpressed in several cancer cells.…”

“…Transplantable B16Fl0 melanoma cells from C57BL/6J mice were obtained from the American Type Culture Collection. Previously, immunohistochemical analysis has verified the abundance of ectopic mGluR1 on the surface of B16F10 melanomas (14,15). The B16Fl0 cells were maintained and passaged in Dulbecco's modified Eagle's medium supplemented with 10% fetal bovine serum, penicillin (100 U/mL), and streptomycin (0.1 mg/mL).…”

“…We previously developed 4-18 F-fluoro-N-[4-[6-(isopropylamino) pyrimidin-4-yl]-1,3-thiazol-2-yl]-N-methylbenzamide ( 18 F-FITM; Fig. 1) to visualize and quantify mGluR1 expression in the brain (12,13) and melanomas (14). This radiotracer is a specific antagonist for mGluR1, with a half maximal inhibitory concentration (IC 50 ) value of 5.1 nM, and displays excellent selectivity for mGluR1 compared with other subtypes (IC 50 .…”

“…21) In brief, 7-µm-thick frozen tissue sections were air-dried, blocked for nonspecific binding in HBSS with 25 mM HEPES and 0.1% BSA for 20 min, and incubated for 1 h in optimized binding buffer (determined by in vitro binding assays) containing PET Imaging and Biodistribution Study An Inveon small-animal PET system (Siemens, Knoxville, TN, U.S.A.) was used for dynamic scanning (30 scans of 1 min each) after injection of the imaging agents via tail vein. Three groups of mice were studied, including 2 groups of B16-F10 tumor-bearing mice administered with 18 F-PR_b (8.3±2.2 MBq, n=12) and 18 F-PR_b control (10±1.9 MBq, n=9), and another group of SW48 tumor-bearing mice administered with 18 F-PR_b (8.6±2.6 MBq, n=5).…”
