Molecular insights into the distinct signaling duration for the peptide-induced PTH1R activation

Abstract: The parathyroid hormone type 1 receptor (PTH1R) is a class B1 G protein-coupled receptor (GPCR) that plays critical roles in bone turnover and Ca2+ homeostasis via mediating the actions of two distinct peptides, PTH and PTHrP. The major difference between the two endogenous peptides or their drugs analog, Teriparatide (PTH) and Abaloparatide (ABL), is their marked duration distinctions of the downstream signaling, a key determinant that is responsible for their distinct physiological or pharmacological propert… Show more

“…While this work was being finalized, Zhang et al and Kobayashi et al made available reports detailing cryo-EM structures of PTH1R bound to PTH and ABL (Liu et al, 2022) and PTH and PTHrP (Kobayashi et al, 2022), respectively. Our structures of PTH, ABL, and PTHrP closely agree with those presented by the other groups, these three reports substantially bolster understanding for the molecular basis of PTH1R signaling.…”
