Molecular mechanisms of EGF signaling-dependent regulation of pipe, a gene crucial for dorsoventral axis formation in Drosophila

Abstract: During Drosophila oogenesis the expression of the sulfotransferase Pipe in ventral follicle cells is crucial for dorsoventral axis formation. Pipe modifies proteins that are incorporated in the ventral eggshell and activate Toll signaling which in turn initiates embryonic dorsoventral patterning. Ventral pipe expression is the result of an oocyte-derived EGF signal which down-regulates pipe in dorsal follicle cells. The analysis of mutant follicle cell clones reveals that none of the transcription factors know… Show more

“…Apart from aforementioned grk, pipe was also not expressed by P. aegeria . Pipe plays an essential role in establishing DV polarity in D. melanogaster oocytes, with its expression being confined to ventral follicle cells as a result of localised EGF signalling [91]. Recently, however, it has been proposed that pipe is not necessary in a number of insect species studied [4] and even in D. melanogaster there appears to be a second mechanism in establishing DV [92] that may involve delayed induction by graded maternal Dpp signalling in the perivitelline space [93].…”

“…Transcription factors that have been discussed as mediators of EGF signalling include pointed ( pnt ), aop and capicua ( cic ) [91]. Only the latter two were expressed by P. aegeria and present as maternal transcripts, but whether they play a role in establishing DV polarity remains to be investigated (Tables 6 and 7, and Additional file 2; qPCR results).…”

“…How does the Torpedo signaling pathway repress pipe transcription? Analyses of the transcriptional regulatory elements controlling pipe expression have identified a stretch of approximately 50 nucleotides about 1.1 kb upstream of the transcriptional start site that is highly conserved in pipe genes from a number of Drosophila species . DNA fragments containing this element direct the transcription of a reporter gene in a pattern that is indistinguishable from that of pipe, and mutations introduced into the conserved 50 bp element lead to de‐repression of reporter gene expression in dorsal follicle cells.…”
