Molecular pathogenesis of bladder cancer

Abstract: Bladder tumors show widely differing histopathology and clinical behavior. This is reflected in the molecular genetic alterations they contain. Much information has accumulated on somatic genomic alterations in bladder tumors of all grades and stages and when this information is related to the common histopathological appearances, a model for the pathogenesis of two major groups of bladder tumors has emerged. This review summarizes the genetic alterations that have been reported in bladder cancer and relates t… Show more

“…In superficial Ta bladder tumours, activation of Ras signalling is caused by gain-of-function mutations in HRAS, KRAS, NRAS and FGFR3. Although these mutations are less frequent in T 2 -T 4 bladder cancers, the RAS pathway seems to be affected in highstage tumours as well as by amplification/overexpression of different RTKs, including ERBB2 and ERBB1 (Knowles, 2008;Mitra and Cote, 2009). Two levels of Fra-1 upregulation via the RAS pathway, transcriptional and protein stabilisation, have been described previously (Casalino et al, 2003;.…”

“…TCCs represent the absolute majority of bladder tumors and are divided into two types, non-muscle invasive (ϳ70%) and muscle invasive (15). In many cases, muscle invasive TCC manifest metastatic features that represent a major cause of death.…”
