Molecular phenotyping of circulating tumour cells in patients with prostate cancer: prediction of distant metastases

Abstract: What's known on the subject? and What does the study add?The role of circulating cancer cells in metastogenesis is generally accepted. Two forms of these cells have been reported in a number of studies, cancer cell clusters (CCCs) and individual epithelial cancer cells. Clusters appear at higher frequencies in the blood. CCCs have been reported to be rich in vimentin and poor in E‐cadherin expression The resulting epithelial to mesenchymal transition, a prerequisite for metastasis formation, occurs in CCCs. We… Show more

“…Several of these pumps, especially the MDR1 gene-encoded P-glycoprotein (P-gp), possess strong affinities for docetaxel [6]. Furthermore, increased expression of MDR1 in circulating tumour cells from a series of patients with prostate cancer has been associated with a decreased response to docetaxel chemotherapy and OS [7].…”

“…GPX1 has largely been studied in relation to carcinogenesis, although transcript levels for GPX1 along with other antioxidants have been suggested to have predictive value for metastasis (Giesing et al, 2012; Han et al, 2013; Miar et al, 2015; Min et al, 2012). Increased GPX1 activity, but not expression levels, was linked to increased vascular invasion using a hepatocellular carcinoma cell model (Huang et al, 2012).…”

“…CTCs have been found in clusters, potentially representing tumor emboli or a product of intravascular proliferation [8,9]. The exact significance of CTC clusters is unclear, though one study has shown their different gene expression profiles when compared to individual CTCs [10]. …”

“…performed molecular phenotyping of PCa CTCs with direct links to clinical outcomes [10]. They tested functional gene targets as biomarkers for disease, analyzing 23 targets and identifying overexpression of five genes (SOD2, GPX1, AR, cyclin B and bFGF) that predicted clinical metastases.…”
