Molecular Physicochemical Properties of Selected Pesticides as Predictive Factors for Oxidative Stress and Apoptosis-Dependent Cell Death in Caco-2 and HepG2 Cells

Abstract: In this work, three pesticides of different physicochemical properties: glyphosate (GLY, herbicide), imidacloprid (IMD, insecticide), and imazalil (IMZ, fungicide), were selected to assess their cytotoxicity against Caco-2 and HepG2 cells. Cell viability was assessed by the Alamar Blue assay, after 24 and 48 h exposure to different concentrations, and IC50 values were calculated. The mechanisms underlying toxicity, namely cellular reactive oxygen species (ROS), glutathione (GSH) content, lipid peroxidation, lo… Show more

“…In addition, the cell types used might be a factor in obtaining different results. In a study with pure glyphosate, however, ROS level was not changed by 250-750 µM concentrations at 24 h [38]. In our study, decline of ROS at higher dose (200 µM) might be related to excessive cytotoxicity at high doses.…”

“…Mitochondrial membrane potential (MMP) was evaluated using 5,5 ′′ ,6,6 ′′ -tetrachloro-1,1 ′′ ,3,3 ′′ -tetraethylbenzimidazolylcarbocyanineiodide (JC-1). Cells were processed as described for DCFDA but incubated with 200 µL of JC-1 (2 µM, in FBS-free DMEM) for 20 min (37 • C, in the dark) [65]. As JC-1 is a ratiometric indicator of MMP variations, the results are presented as the ratio of mean FL2/FL1 fluorescence intensities, normalized by the negative control (nonexposed cells).…”

“…Concerning IMD, this insecticide was quantified in many food products, and a daily intake of IMD was estimated to be 0.004-0.131 µg/kg body weight [50]. According to cell-based assays, besides dose-dependent toxicity, cytotoxicity is also dependent on the pesticide, the cell type and exposure time [51,52]. In order to compare the effects of the four pesticides, the same range of concentrations (up to 1 mM) was used in the experiments.…”
