Molecular sex determination of 20 bird species protected in the Republic of Serbia

Abstract: Considering that more than 50% of bird species are monomorphic, the identification of gender based on phenotypic characteristics is extremely difficult. The aim of this study is sex determination in species inhabiting the Republic of Serbia, all under the state protection and declared by IUCN as endangered. DNA was isolated from feathers. Sex determination was based on sex-specific CHD gene amplified by 2550F/2718R primer set. Sexing gave good results in all 91 samples from 20 species including 6 species… Show more

“…When it comes to sex determination in sampled birds using a dye-labelled CHD1F/CHD1R primer set, capillary electrophoresis proved to be an accurate, fast, and reliable method. Our results confi rmed that the specifi ed method enabled accurate sex determination in all tested species, although end point PCR approach was also highly successful, as shown here, but also in previous studies [7,9,10,26,27].…”

“…To reduce wildlife traffi cking and illegal hunting, international organizations and national agencies highly depend on forensic investigations which include species and sex identifi cation of animals [7][8][9]. In the last decades, molecular techniques have become very valuable forensic tools that enable, among other things, identifi cation of species from a very low quantity of samples [2,10,11] of various types (e.g.…”

“…In that experiment, C-banding was performed using the peripheral blood lymphocyte division phase, and the W chromosome was successfully identified, allowing the sex of the early embryos of intergeneric hybrids to be determined. In the present study, we used C-banding to identify W chromosomes, in combination with the morphological identification of Z chromosomes, which greatly improved the accuracy of sex determination of birds, providing a safe and reliable sex identification test for certain rare avian species [21,22] .…”
