Molecular Tuning of the Nano–Bio Interface: Alpha-Synuclein’s Surface Targeting with Doped Carbon Nanostructures

Abstract: Carbon nanoparticles are becoming promising agents in treating Parkinson's disease (PD) by preventing the folding and aggregation of α-synuclein, i.e., amyloid formation. Herein, for the first time, highly tunable graphene and carbon nanotubes (CNTs) have been doped using biocompatible silicon atoms for preventing Parkinson's disease. In this study, the conformational changes induced by these nanoparticles, the compactness of nanoparticles, the number of hydrogen bonds, the stability of α-synuclein in the pres… Show more

“…Modifying the surface characteristics of nanoparticles, such as with Polyethylene Glycol (PEG) modifications, can create "stealth" nanoparticles, 44,141 impeding the Reticuloendothelial System (RES) from phagocytosing drugs entering the bloodstream. [142][143][144] This modification significantly extends the drug's half-life in the blood. The primary intracellular barrier during delivery is the endosome/lysosome system.…”

“…Significantly, these nanoparticles affected the secondary structures of α-syn, reducing the β-sheets forms that support their capability to prevent the folding and aggregation of α-syn. 51 Based on the MD results by Mohammad-Beigi and co-workers (2019), positively and negatively charged graphene sheets interact with terminally charged α-syn N/C residues and then with hydrophobic residues (61–95) of the non-amyloid-β component region driving α-syn fibrillation. Accordingly, graphene sheets not only inhibited α-syn fibrillation (both nucleation and elongation), but even disrupted the mature fibrils.…”
