Monitoring of intracellular localization of Hepatitis B virus P22 protein using Laser Scanning Confocal Microscopy and Airyscan

Abstract: The aim of this study was to assess nucleo-cytoplasmic protein localization to better understand the exact intracellular localization of viral proteins involved with infections. Having determined the general protein localization of hepatitis B virus P22 precore protein, the aim was to more specifically resolve its intracellular organization. This was done using both laser scanning microscopy and Airyscan techniques. Using a 63× objective, the resolution obtained with Airyscan was increased by 1.5-fold as compar… Show more

“…There was increased retention of viral proteins in the secretory pathway, increased ER stress, and earlier and prolonged activation of UPR in cells transfected with subgenotype A1 [44]. When liver cells were transfected with replication competent plasmids of subgenotype A1, precore protein expression was affected by absence of core, which also influenced HBsAg expression, suggesting an interrelationship between precore proteins, HBcAg, and HBsAg [65]. Incorporating greater-than-genome-length subgenotype A1 sequences into recombinant adeno-associated viruses (AAVs), HBV replication was demonstrated in vitro in hepatoma cells and in mice [66].…”

“…For size distribution estimation, bright-field microscopy images were captured of a 0.01% dilution of Zuper Black using the bright-field component of a ZEISS LSM 780 confocal laser scanning microscope with an alpha Plan-Apochromat 63×/1.40 NA Oil CorrM27 objective lens (Carl Zeiss AG, Oberkochen, Germany) [39].…”

“…This technique, called laser scanning confocal microscopy (LSCM) [ 8 , 9 ], was originally developed for application in biology and medicine. Now, it is more and more widely used and has become an essential tool in biomedical experimental research [ 10 , 11 , 12 , 13 ]. Compared to conventional fluorescence microscopy, it overcomes the shortcomings of an out-of-focal plane-fluorescence imaging blur and focal plane glare and can improve the optical resolution of fluorescence microscopy.…”
