Monitoring Seasonal Changes in Winery-Resident Microbiota

Abstract: During the transformation of grapes to wine, wine fermentations are exposed to a large area of specialized equipment surfaces within wineries, which may serve as important reservoirs for two-way transfer of microbes between fermentations. However, the role of winery environments in shaping the microbiota of wine fermentations and vectoring wine spoilage organisms is poorly understood at the systems level. Microbial communities inhabiting all major equipment and surfaces in a pilot-scale winery were surveyed ov… Show more

“…Amplification and sequencing were performed as described previously for analysis of bacterial (26) and fungal communities (53). Briefly, the V4 domain of bacterial 16S rRNA genes was amplified using primers F515 (5′-NNNNNNNNGTGTGCCAGCMGCCGCGG-TAA-3′) and R806 (5′-GGACTACHVGGGTWTCTAAT-3′) (54), with the forward primer modified to contain a unique 8-nt barcode (italicized poly-N section of primer above) and a 2-nt linker sequence (bold portion) at the 5′ terminus.…”

“…However, recent advances in massively parallel, short-amplicon sequencing technologies have launched a breakthrough in microbial ecology studies of wine-and foodfermentation systems (14,(21)(22)(23)(24)(25)(26)(27), putting hitherto untenable ecological questions within reach.…”

“…Moreover, the interplay between the wine microbiota and the microbiota of the fermentation facilities has been verified but not completely understood [6]. Due to this complexity of interactions between microorganisms in the wine itself, during fermentation, but also between wine microbiota and environment, the wine industry has adopted the use of starter cultures as a mean of control and quality improvement [7].…”

“…Using these culture-based techniques, Vacheyrou previously examined possible routes of microbial transfer in farms supplying raw milk for Comte style cheese, revealing that the extent to which milk was contaminated varied depending on the type of barns used to house animals (2). However, recent advances in molecular microbiology, and in high-throughput DNA sequencing (HTS) in particular, have allowed for a more in-depth analysis of the flow of microbes through environments (7)(8)(9)(10)(11)(12).…”
