Monoclonal Antibody Against a Melanosomal Protein in Melanotic and Amelanotic Human Melanoma Cells

Abstract: BALB/c mice were immunized with tyrosinase, partially purified in two stages from a human melanoma cell line. A hybridoma was obtained which produced monoclonal antibody (MoAb 1C11) reactive with 8/10 melanoma cell lines and 10/10 primary cultures of human melanocytes, neval cells, and melanomas. Immunoreactivity correlated to a certain extent with tyrosinase activity but not with melanin content. No crossreactivity was obtained with neuroblastoma, medulloblastoma, fibroblasts, keratinocytes, lymphoid cells, o… Show more

“…Color was developed by reaction with 50 pl of 7.5 mM BCIPl0.7 mM NBT at room temperature for 30 min. Positive staining with both the rabbit anti-bovine SlOO antibody (DAKOPA'M'S; Nakajima et al, 1982) and the monoclonal antibody l C l l to a melanosomal antigen (McEwan et al, 1989) confirmed the identity of cells as melanoma.…”

“…Several studies have reported the development of murine MoAbs against human melanosomes by immunising animals with intact human melanoma cells or with melanosomes derived from these cells (McEwan et al, 1989;Hayashibe et al, 1986). Four MoAbs, HMSA-1 to 4, previously developed by the latter method in this laboratory (Akutsu & Jimbow, 1986;Jimbow et al, 1990) were shown to recognise melanosomal antigens highly specific to neoplastic melanocytes.…”

“…MoAbs l C l l and 5C12 react with human melanosomal antigens and have been described previously (McEwan et al, 1988(McEwan et al, ,1989. MoAb 2B7 binds active tyrosinase but appears to recognise a different epitope from MoAb 5C12 (unpublished).…”
