Monoclonal Antibody Production and the Development of an Indirect Competitive Enzyme-Linked Immunosorbent Assay for Screening Spiramycin in Milk

Abstract: To monitor spiramycin (SP) residue in milk, a monoclonal antibody (mAb)-based indirect competitive enzyme-linked immunosorbent assay (icELISA) was developed. This study described the preparation of three immunogens and the production of a high-affinity mAb. After optimization, the 50% inhibition concentration (IC50) for the developed icELISA was estimated as 0.97 ng/mL in the assay buffer, and the limit of detection and limit of quantitation were 2.51 and 4.40 μg/L in the milk matrix. The newly developed assay… Show more

“…After an elaborate antibody validation benzoylecgonine, MDMA and methadone were detectable and quantifiable specifically and sensitively. Comparing the results to other competitive immunoassays for substances like antibiotics (Huang et al, 2010;Jiang et al, 2013;Wang et al, 2015), similar detection limits and CVs were obtained. In relation to one of the standard application of immunoassays for drug abuse (lateral flow assays) better detection limits and quantification could be reached (Attema-de Jonge et al, 2012;Taylor and Pizzo, 2004).…”

“…Hapten and antigen synthesis The hapten was synthesized using the CMO method (Figure 2) (Jiang et al, 2013). Onehundred mg of KIT and 20 mg of CMO were added to a mixture of 5 mL methanol and 5 mL 0.02 M sodium bicarbonate solution.…”

“…The analytical performances of ELISA are highly dependent on the activity of antibody and enzyme label. In biomedical, feed/food and environmental settings, many chemicals, such as toxins, antibiotics and some synthetic dyes, are inspected through ELISA methods (Cao et al, 2013;Du et al, 2016;Jiang et al, 2013;Peng, Duan, Khamba, & Xie, 2014;Wang, Li, Zhang, Hu, & Zhang, 2016;Xu et al, 2012). Nevertheless, the analytical performances of the conventional ELISA methods are needed to be improved in order to facilitate their application in practice (Lei & Ju, 2012).…”
