Monoclonal antibody stains oligodendrocytes and Schwann cells in zebrafish ( Danio rerio )

Abstract: We prepared a monoclonal antibody that recognizes oligodendrocytes and Schwann cells in zebrafish. On immunoblots, the antibody mainly recognized three protein bands of 34 kDa in a membrane fraction from adult zebrafish brain. Medaka fish (Oryzias latipes) also possessed the same protein bands in a membrane fraction. The antibody did not stain neurons, but stained cells in fiber tracts and cranial and spinal nerves. In order to determine the nature of these cells, the staining pattern of the monoclonal antibod… Show more

“…Synaptic membranes from zebrafish brain were prepared by the method used to prepare bovine brain synaptic membrane fractions (Tomizawa, 2000a and2000b). Zebrafish were maintained at 28.5 o C under a 14h light and 10h dark cycle (Westerfield, 1995).…”

“…Briefly, a zebrafish head was fixed in FX buffer (Tomizawa et al, 2000a), containing 4% freshly depolymerized paraformaldehyde at 4°C overnight. The brain was carefully removed from the skull and gently washed in 0.1 M sodium phosphate buffer (pH 7.3) containing 4% sucrose and 0.15 mM CaCl 2 and then placed into a 30% sucrose solution until it sank.…”
