Monoterpenes in the glandular trichomes of tomato are synthesized from a neryl diphosphate precursor rather than geranyl diphosphate

Abstract: We identified a cis-prenyltransferase gene, neryl diphosphate synthase 1 (NDPS1), that is expressed in cultivated tomato (Solanum lycopersicum) cultivar M82 type VI glandular trichomes and encodes an enzyme that catalyzes the formation of neryl diphosphate from isopentenyl diphosphate and dimethylallyl diphosphate. mRNA for a terpene synthase gene, phellandrene synthase 1 (PHS1), was also identified in these glands. It encodes an enzyme that uses neryl diphosphate to produce ␤-phellandrene as the major product… Show more

“…Terpenes are stored in glandular trichomes of Solanum lycopersicum leaves and are released even in physiological conditions at a level of 0.5 nmol m -2 s -1 which is in accordance with data obtained by Schimiller et al [32,33]. The emissions consisted of α-pinene, camphene, 2-carene, α-phellandrene, limonene, β-phellandrene, (E)-β-ocimene and terpinolene as have been previously reported by Jensen et al [21] and Copolovici et al [16].…”

“…The condensation reactions initially form geranyl diphosphate (GPP) and farnesyl diphosphate (FPP), which can be modified by terpene synthases to produce over 1000 monoterpenes and approximately 5000 sesquiterpenes, respectively [55][56][57][58][59][60][61]. Monoterpenes are derived from GPP by the action of monoterpene synthases, and sesquiterpene synthases transform FPP to various sesquiterpenes [62]. An overview of the enzymatic reactions resulting in synthesis of major lavender monoterpenoids is shown in l " Fig.…”

“…Likewise, the overexpression of the lemon basil α-zingiberene synthase gene in tomato plants resulted in the production of a monoterpene in the cytosol (Davidovich-Rikanati et al 2008). Santalene and bergamotene synthase (SBS) from tomato contain putative N-terminal plastid targeting peptides (Schilmiller et al 2009). Plant genomes have evolved to encode diverse FPP synthase isoforms, which are present in the cytosol, mitochondria, plastids or peroxisomes Thabet et al 2012).…”
