Morphogenetic responses from in vitro cultured seedling explants of mung bean (Vigna radiata L. Wilczek)

Abstract: The morphogenetic responses of seedling explants of mung bean (Vigna radiata L.Wilczek cv ML-5) were studied in vitro. Direct induction of shoots/plants was possible from shoot tip, cotyledon and cotyledonary node explants. Dedifferentiation of the explants viz; Shoot tip, cotyledons, cotyledonary node, primordial leaves and roots was obtained on basal medium supplemented with auxin and cytokinin. Shoot regeneration was limited to primary calli while rhizogenesis was of common occurrence in established calli. … Show more

“…Regeneration from various seedling explants viz shoot tip, cotyledons, cotyledonary node of mung on BAP supplemented medium has been reported earlier (Mathews, 1987;Gulati & Jaiwal, 1992;Kaviraj & Rao, 2009;Mendoza, et al, 1999).there is only one report of regeneration from callus cultures in mungbean , similarly plantlets have been regenerated from NaCl tolerant callus in many species like Citrus sinensis (Ben-Hyyim & Goffer,1989) Brassica juncea (Jain, et al,1991,Kerthi, et al, 1989Sashimadan, et el., 1995) Triticum durum (Zair, et al, 2003) Ipomea batats (He, et al, 2009) In the present study however, it was noticed that the formation of shoots from NaCl tolerant callai was lower (14.00% than that from NaCl-non selected calli (40.13%) and the number of multiple shoots per callai piece was reduced to 3.33 from 14.80, a phenomenon reported earlier by few workers (Basu, et al, 1997;Chen et al, 2001;Ochatt, et al,1999;Queiros, et al, 2007;Zair et al, 2003).…”

“…Par ailleurs, la kinetine a favorisé l'induction des cals mais à partir de la mise en culture in vitro des ovules du mandarinier (Handaji et al , 2005, Singh et al 2006. La nature, l'âge, la position, l'orientation des explants, leurs besoins spécifiques en éléments nutritifs et leurs teneurs endogènes en hormones influencent la réponse à la callogenèse (Mathews, 1987 Gmitter et Moore, 1986et Bugam et al 2003, Handaji et al, 2005. “…13 In vitro cultures of Vigna radiata were reported earlier. [14][15][16][17] The present research investigated the biotransformation possibility of hydroquinone to arbutin for the first time. On the other hand, it evaluated its bioconversion capacity by adding elicitors like chitosan and methyl jasmonate to V. radiata cell culture.…”
