“…In this study, we link the activity of WNT signaling to the tumorigenic potential of ESC-RPCs and provide the experimental evidence for transcriptional factor TCF7 to regulate expression of SOX2 and NESTIN, two important genes actively engaged in the neural development and tumor formation. The tumorigenic and therapeutic effect of transplanted ESC-RPCs is determined in a well-studied sodium iodate-induced (SI-induced) mouse retinal degeneration model (20,21). We also show that the expression of TCF7, SOX2, and NESTIN is closely associated in mouse neonatal retinae.…”

“…The oxidative stressor sodium iodate (NaIO 3 ) has been used previously to induce selective degeneration of the RPE and consequent retinal degeneration (Ringvold et al 1981;Kiuchi et al 2002;Enzmann et al 2006). Intravenous injection of NaIO 3 into C57/BL6 mice induced disorganization of the RPE layer after 36 h, and subsequently led to the degeneration of photoreceptors.…”

“…NaIO 3 injection and RPE lysate preparation C57/BL6 mice (2 to 3 mo) were injected with NaIO 3 solution (30 mg/kg in PBS) into tail veins as described previously (Kiuchi et al 2002;Enzmann et al 2006). Eyes were enucleated and the retina carefully pushed out before treating with dispase (100 U/mL) to isolate RPE cells.…”

“…On the other hand, the majority of animals did not show an increase but rather a further decrease of their visual function. This might be due to the already initiated apoptosis in the photoreceptors that follows the initial damage of the RPE by NaIO 3 [29] . The impact of the subretinal surgery on the visual function should also be taken into account.…”
