MR Imaging Characterization of Microvessels in Experimental Breast Tumors by Using a Particulate Contrast Agent with Histopathologic Correlation

Abstract: USPIO, a macromolecular particulate MR imaging contrast agent, can be applied successfully to characterize tumor microvessels in animals. USPIO-derived K(PS) correlated strongly with histopathologic tumor grade, MVD, and K(PS) values derived by using albumin-(Gd-DTPA)(30) in the same tumors.

“…By comparison, the blood pool agent albumin(Gd-DTPA) 30 is largely retained in the intravascular space with a half-life of 88min after intravenous injection and can remain in the bloodtstream and extravascular compartment two weeks after injection. (77,78) These undesirably long retention times have precluded its use in the clinic.…”

“…68 In addition, superparamagnetic contrast agents such as iron oxide particles (ultrasmall superparamagnetic iron oxide) successfully determined blood volume and tumor vasculature on the basis of the phagocytic activity of macrophages. 69 Nevertheless, the evaluation of plaque neovessels with the use of MRI-ultrasmall superparamagnetic iron oxide has not been published yet.…”

“…Reflux k is commonly set to zero in DCE-MRI studies that use large-molecular contrast agents [7,16]. In our study, k was calculated, but no conclusions were drawn that pertained to differences between treatment groups or differences within a tumour.…”

“…This region of interest was used for further analysis. Regions of interest for the tumour rim were defined as the approximately 1 to 2 mm-thick peripheral zone of strong contrast enhancement [7] in the dynamic T 1 -weighted series; the tumour core was the remaining central region.…”
