mRNA COVID-19 Vaccines—Facts and Hypotheses on Fragmentation and Encapsulation

Abstract: Background: The adventure of the mRNA vaccine began thirty years ago in the context of influenza. This consisted in encapsulating the mRNA coding for a viral protein in a lipid particle. We show how the mRNA encoding S protein has been modified for that purpose in the context of the anti-SARS-CoV-2 vaccination. Results: by using data coming from genetic and epidemiologic databases, we show the theoretical possibility of fragmentation of this mRNA into small RNA sequences capable of inhibiting important bio-syn… Show more

“…Hence, for the USA and India, a minimum of the ID variable was impossible to individualize inside the endemic background noise. The future efforts in the direction of this research are vital for a future pandemic or emerging infectious disease preparation, because we believe that the research presented in this article could be relevant for new infectious case forecasting in order to deploy proper intervention and resources (as vaccination policies [ 48 ]) to fight the epidemic spread and, in a way, guide policy-making for public health.…”

“…This process is comparatively straightforward when compared to the bioengineering challenges associated with distinct proteins or peptides. For example, the sequence identification and optimization of COVID-19 mRNA vaccines have been intensively explored [ 98 ]. Additionally, the mRNA-1653 vaccine exemplifies the capability of a single vaccine to simultaneously target two distinct pathogens.…”

“…Similarly to SARS-CoV-2 mRNA, synthetic mRNAs exhibit quite unique behavior related to their chemical structure, nucleotide sequence, and formulation. Interestingly, their concentration, stability and distribution on tissues, organs and cells also occur through genome integration via L1 elements [61][62][63][64]. This reflects either the amount of vaccine mRNA delivered in a single dose, the presence of SNPs that may impair the IFN-a (major somatic cell antiviral defense mechanism) expression, or the defection of the antiviral RNA interfering mechanism ( RNAi) typical of multipotent and pluripotent SCs [65][66][67][68].…”
