MT1-MMP sheds LYVE-1 on lymphatic endothelial cells and suppresses VEGF-C production to inhibit lymphangiogenesis

Abstract: Lymphangiogensis is involved in various pathological conditions, such as arthritis and cancer metastasis. Although many factors have been identified to stimulate lymphatic vessel growth, little is known about lymphangiogenesis inhibitors. Here we report that membrane type 1-matrix metalloproteinase (MT1-MMP) is an endogenous suppressor of lymphatic vessel growth. MT1-MMP-deficient mice exhibit spontaneous corneal lymphangiogenesis without concomitant changes in angiogenesis. Mice lacking MT1-MMP in either lymp… Show more

“…One example is via suppression of vascular endothelial growth factor C (VEGF-C) expression, mediated in part by membrane type 1-matrix metalloproteinase signaling that suppresses macrophage production of VEGF-C 41 . Likewise, the matricellular protein thrombospondin 1 (TSP1) also suppresses macrophage expression of VEGF-C, as described by Masli and colleagues 42 .…”

“…The processing converts EphA2 into a ligand‐independent signal transducer that shows opposite activity of the ligand‐bound receptor by promoting cell motility . MT1‐MMP also sheds CD44 and its homolog lymphatic vessel endothelial hyaluronan receptor 1 and enhances cell motility and lymphangiogenesis, respectively . Likewise, biological feedback due to MT1‐MMP expression in producer cells reflects the altered functions of the processed substrate proteins.…”

“…(5,6) MT1-MMP also sheds CD44 and its homolog lymphatic vessel endothelial hyaluronan receptor 1 and enhances cell motility (7) and lymphangiogenesis, respectively. (8) Likewise, biological feedback due to MT1-MMP expression in producer cells reflects the altered functions of the processed substrate proteins. Therefore, the diverse functions of MT1-MMP in tissues reflect the available substrates in the cellular microenvironment.…”

“…In a recent study, analysis in mice mutants for MMP14 (Matrix Metalloproteinase 14), also known as MT1‐MMP, revealed a vast increase in corneal and diaphragmatic lymphangiogenesis marked by LYVE1 staining . Increased lymphangiogenesis was accompanied by upregulation of Vefgc and Vegfd but not Vegfa , supporting a lymphatic rather than angiogenic effect.…”
