-DNA sequence diversity in a non-coding portion of the mitochondrial genome was investigated in samples of Apis cerana from 47 locations in China. Nine haplotypes (mitochondrial genotypes) were found: Japan1, Japan2, Korea4, and Cambodia2, which were previously reported from other populations, and China1−5, which are new. All nine sequences belong to the Mainland mitochondrial lineage, and none differs from the Japan1 haplotype by more than a single base substitution and/or a single insertion/deletion. Japan1 is the most common haplotype, making up 39 of 49 sequences. Haplotype diversity was 0.4 (s.d. 0.089) and nucleotide diversity was 0.00569 (s.d. 0.00154). By both measures the Chinese samples were more diverse than those from Japan and Thailand, similar to populations from Pakistan, Burma and Korea, and less diverse than samples from Indochina (Laos-Cambodia-Vietnam).mitochondrial DNA / Apis cerana / China / phylogeography

Morphometric analysis of Apis cerana populations in the southern Himalayan region

-Multivariate analyses of morphometric traits of Apis cerana were measured from 3704 individual workers of 279 colonies from 64 localities with an average sampling distance resolution of 50 km along a 2200 km transect in the southern Himalayan region bordered by Pakistan to the west and Myanmar to the east. Factor and discriminant function analyses revealed four major morphoclusters (= unnamed subspecies), two of which are further subdivisable into three biometric subgroups each. These morphoclusters can only be partially integrated into any current subspecific nomenclature available for Apis cerana. Morphocluster separation is related to physiographic differences which create a partial temporal reproductive isolation associated with altitude. High variance domains occur at the edges of morphocluster and biometric groupings. The bees decrease in size from west to east but increase in size with increasing altitude. ABSTRACT. A molecular maker for authenticating species origin of the stingless bee (Trigona collina) was developed. Initially, amplified fragment length polymorphism analysis was made of 11 stingless bee species using 64 primer combinations. A 316-bp band found only in T. collina was cloned and sequenced. A primer pair (CUTc1-F/R) was designed and tested for species-specificity in 15 stingless bee species (239 nests). The expected 259-bp fragment was consistently amplified in all T. collina individuals (134/134 nests, 100%). Cross-species amplification was observed in T. pagdeni (43/51 nests; 84.3%), but not in other species. SSCP analysis of CUTc1 unambiguously differentiated T. collina from T. pagdeni. CUTc1 generated three genotypes in Thai T. collina (134 nests). An AA (259/259 bp) genotype was found in all stingless bees from the north (21 nests) and northeast (32 nests), and 23/28 nests from the Central region, whereas a BB (253/253 bp) genotype was observed in most samples from peninsular Thailand (42/53 nests). Heterozygotes exhibiting the AB (253/259 bp) genotype were observed in 5 of 28 nests from Prachuap Khiri Khan located slightly above the Kra ecotone and 11 of 53 nests originated further south of the Kra ecotone. Genotype distribution patterns of CUTc1 clearly indicated intraspecific population differentiation of Thai T. collina.
