Mucoepidermoid tumors of salivary glands: histoprognostic value of NORs stained with AgNOR technique

Abstract: A total of 31 patients surgically treated for salivary mucoepidermoid tumor had a comprehensive follow-up. Two groups were classified as: 16 with favorable outcome (no recurrence or late recurrences at 5 yr postoperatively and still alive; 15 with poor outcome (local recurrences before 5 yr, metastases, lethal evolution in 11 cases). We evaluated nucleolar organizer in every case, using AgNOR count. The AgNOR count seemed better than histologic criteria for establishing the prognosis of mucoepidermoid tumors. … Show more

“…PCNA immunostaining and AgNOR staining have been established as valuable methods with which the biological behavior of malignant cells can be examined [Robbins et al, 1987;Chomette et al, 1991;Tsuji et al, 1992;Cabrini et al, 1992]. PCNA was detected as a nuclear antigen reacting to autoantibodies in sera o f patients with systemic lupus eryth ematosus [Miyauchi et al, 1978], and is described as a non histone nuclear protein that appears in the nucleus during the late G1 phase, increases during the S phase and declines during G2 and M phases of the cell cycle [Celis and Celis, 1985].…”

“…Due to its simplicity and sensitivity, its rapid and convenient nature (6)(7)(8)(9)(10)(11), and its applicability to paraffin sections, argyrophillic nuclear organizer region (AgNOR) analysis was used in the present study. Nucleolar organizer regions (NORs) are loops of DNA that encode ribosomal RNA and are considered to be important in the synthesis of proteins.…”

“…The grading system for MEC reported by Seifert et al, which is based on proportion of cells in a given section, is less reliable than AgNOR count in MEC in the context of MEC outcome (6). Auclair et al described a threetiered grading system based on various histopathological features ( Table 1).…”

“…Thus, this method has become an important auxiliary tool for the anatomo‐pathological diagnosis (6, 12, 13). Likewise, it has been used to diagnose oral diseases, such as salivary glands tumors (4, 14, 15), odontogenic cysts, odontogenic fibroma, ameloblastoma (16) and different lesions that affect the oral mucus, generally of cancerous development (17–21).…”
