Multi‐component profiles through the blood–brain barrier in rat after oral administration of over‐the‐counter drug Keke capsule by ultra‐performance liquid chromatography/quadrupole‐ time‐of‐flight MSE method

Abstract: Keke capsule as a traditional Chinese medicine formulation is used to relieve cough, for analgesia and to reduce bronchial asthma. The multi-components are absorbed into the blood and brain after oral administration of Keke capsule, with no systematic investigation so far. A reliable and rapid UPLC-QTOF-MS combined with a data processing software platform was used to characterize the components of Keke capsule and simultaneously identify bioactive components in blood and brain tissues in rat after oral adminis… Show more

“…Mass spectrometric data acquisition is divided into data-dependent acquisition and data-independent acquisition (DIA), where the DIA technique does not require preselection of compounds in the sample but directly acquires the mass spectral information of all compounds separated by the column [7]. As one of the typical DIA techniques, full information MS/MS (MS E ) is a data acquisition method that enables alternating "low collision energy" and "high collision energy" scans to obtain highly accurate information on precursor and fragment ions and enables correlation attribution based on the chromatographic behavior of the precursor and fragment ions [8]. Due to the large differences in content, structural types, and physicochemical properties among the many components of TCM in the qualitative analysis process, in the past, researchers often manually extracted peaks and identified compounds one by one based on information such as quasi-molecular ions, characteristic fragment ions, retention time (Rt), and so forth.…”

“…Ultra-high performance liquid chromatography tandem time-offlight mass spectrometry (UPLC-QTOF-MS/MS) technology combined with information dependent acquisition (IDA) can simultaneously obtain high-quality and accurate primary and secondary mass spectrometry information of the compound. [20] According to the primary and secondary mass spectrometry information, the target compound can be quickly screened or inferred. [21] Since compounds with the same structure have similar fragmentation rules and characteristic fragments in mass spectrometry, the study of the fragmentation rules of similar reference materials and their characteristic fragment ions as a diagnostic basis is conducive to the analysis of the mass spectra of unknown compounds.…”

“…In this study, a fast and effective method was used to analyze the chemical components in HP. Ultra‐high performance liquid chromatography tandem time‐of‐flight mass spectrometry (UPLC‐QTOF‐MS/MS) technology combined with information dependent acquisition (IDA) can simultaneously obtain high‐quality and accurate primary and secondary mass spectrometry information of the compound [20] . According to the primary and secondary mass spectrometry information, the target compound can be quickly screened or inferred [21] .…”

“…In our previous research, it was found that alkaloids in EP could rapidly cross the blood-brain barrier of rats after oral administration, presenting the different characteristics of regional distribution in 6 brain regions mentioned above. Especially, the cortex and hippocampus were the main distribution regions of alkaloids [27][28][29]. However, whether the specific distributions of EP cause the different changes of metabolism in brain regions is still unknown.…”
