Multi-gene Expression-based Statistical Approaches to Predicting Patients’ Clinical Outcomes and Responses

Abstract: Gene expression profiling technique now enables scientists to obtain a genome-wide picture of cellular functions on various human disease mechanisms which has also proven to be extremely valuable in forecasting patients' prognosis and therapeutic responses. A wide range of multivariate techniques have been employed in biomedical applications on such expression profiling data in order to identify expression biomarkers that are highly associated with patients' clinical outcome and to train multi-gene prediction … Show more

“…As an illustration of the data-driven approach, recently proposed systematic data-driven approaches based on in vitro-generated predictive profiles using cell-line models entail five key technical steps: 1) data collection, 2) quality control, 3) identification of candidate gene biomarkers, 4) construction of a multivariate prediction model, and 5) independent validation of the prediction model (Figure 2) [53-57]. …”

“…The expression microarray technique is a high‐throughput genomic method in systems biology for investigating genome‐wide transcriptional patterns [Cheng et al., ]. The basic idea of expression microarrays is to use fixed DNA segments (probes) to assess the presence and concentration of their complementary RNA targets in a biological sample of interest.…”

“…Also several powerful genome-wide detection tools such as genotyping micro arrays and tiling microarrays have been developed based on genetic information from the Human Genome Project. Nowadays, the analysis of huge amount of data from these high throughput tools is still very challenging [3].…”
