Multi-NSAID-based Zn(ii) coordination complex-derived metallogelators/metallogels as plausible multi-drug self-delivery systems

Abstract: Metallogelators/metallogels derived from a series of multi-NSAID based Zn(II)-coordination complexes displaying anti-cancer and anti-bacterial properties were designed based on a structural rationale as plausible multi-drug self-delivery system.

“…Self-assembly-directed supramolecular gelation with its innovative insight delivers a valuable platform for advanced future technologies. [7][8][9] Assimilation of metal components into gel scaffolds offers an effective approach to advance soft materials with beneficial functionalities, such as electrical and thermal conductivity, [10][11][12][13][14] optical properties, 15,16 stimuliresponsive magnetism, [17][18][19][20] self-healing behavior, 16,17,[21][22][23] catalytic action, [24][25][26][27] antibacterial property, [28][29][30][31][32] sensing, [33][34][35][36][37][38][39][40] and redox properties. 35,41,42 Besides these added benefits, metal incorporation establishes added interactions within the building blocks that help to trigger gelation, [43][44][45][46][47] modifying morphological patterns, 48,…”

“…For example, supramolecular metallogels derived from NSAID derivatives as biomaterials having anticancer and antibacterial properties have been reported. 12,36,37 Interestingly, metallogels derived from nondrug ligands exhibiting "druglike" function have also been reported. 38−41 With this background in mind, we set out to synthesize a series of coordination polymer-based metallogels derived from a terpyridyl ligand (L) equipped with nitrile functionality.…”

“…Indeed, supramolecular biomaterials which have been developed by exploiting various specific, dynamic, and tunable noncovalent interactions are some of the most sought after materials in the 21st century. Innumerable supramolecular materials for biomedical applications such as regenerative medicine, tissue engineering, mimicking of enzymes, medical diagnostics, immunosuppressive materials, anticancer agents, 3D cell cultures, wound healing, biomineralization, antibacterial agents, etc., are being developed. Supramolecular biomaterials are modular; they offer improved therapeutics because of various advantages and opportunities such as quantitative and controlled encapsulation and release of therapeutics, physiologic stimuli-responsive delivery, multidrug-delivery, molecular insights into the degradation mechanism of the delivery platform, etc.…”

“…In this approach, the drug is delivered to the target site by itself (hence the name SDD). It can be achieved by converting the drug/drug derivatives to supramolecular gels 41–46 or nano-sized particles. 47 While supramolecular gels are amenable for topical (non-invasive) or subcutaneous (invasive) application allowing slow and sustained release of the drug, nano-sized particles can easily penetrate the cell wall, thereby enabling its delivery to the target site.…”
